Oligonucleotide mit antiviralen Nucleosiden
 

Nucleosid-Analoga spielen als antiviral bzw. zytostatisch wirkende Substanzen eine wichtige Rolle. In diesem Zusammenhang sind auch Oligonucleotide mit entsprechenden Nucleosidbausteinen von Interesse, beispielsweise als Prodrugs.
Nachfolgende Abbildung zeigt eine Auswahl von modifizieten Nucleosiden, die in Oligonucleotide eingebaut werden können: 




1: 5-F-dU (5-Fluorouridine, Floxuridine), 2: 5-I-dU (5-Iodouridine, Iodoxuridine), 3: ß-L-dT (ß-L-2'-Deoxythymidine, Telbivudine ), 4 : Ara-A (Arabinoadenosine, Vidarabine), 5: Ara-C (Arabinocytidine, Cytarabine), 6 : dFdC (2',2'-Difluoro-2'-deoxycytidine, Gemcitabine), 7: ddC (2',3'-Dideoxycytidine, Zalcitabine ), 8: 3TC (Lamivudine),  9: Ganciclovir

5-F-dU, 5-I-dU, ß-L-dT, Ara-A, Ara-C, dFdC können sowohl am 3'- oder 5'-Ende als auch in der Sequenz eines Oligonucleotids eingebaut werden, auch mehrfache Kopplungen sind möglich. ddC und 3TC können aufgrund ihrer Struktur lediglich als 3'-Baustein dienen; Ganciclovir bieten wir derzeit ebenfalls nur als 3'-Modifikation an.
